891 Effect of antigen presenting element gene methylation on psoriasis

To explore the relationship between protein expression of antigen presentation related genes (TAP1, TAP2, LMP2, LMP7) and psoriasis their differences. Collect skin tissue samples from patients with normal people, was detected by immunohistochemistry SP method Western blotting, respectively. RT-PCR showed that there no significant difference in TAP1, TAP2 LMP2 healthy controls, LMP7 mRNA decreased significantly. blot compared TAP1 psoriatic lesions significantly increased, had change, reduced. Immunohistochemistry detection number positive cells patients' higher than control group, lower group. Methylation results methylation level controls at 125 locus gene (p<0.05). The increase levels may be to occurrence psoriasis, decrease inhibition antigen-presenting proliferation abnormal Kc further tumor like cells. promoter region APM member can inhibit expression, affect assembly HLA-I molecules, cause down-regulation or deletion on surface keratinocytes, make escape immune surveillance killing body, lead which epigenetic mechanism psoriasis.
